Does NMN Work? What 12 Human Studies Actually Found

A researcher reviewing printed studies

You have seen the NMN headlines. So we did the slow part: we read the human studies, not the mouse ones, and wrote down what each one found and what it did not.

In 30 seconds: NMN is a building block the body turns into NAD+, a molecule cells use to make energy. In every human trial that measured it, NMN raised blood NAD+, and short studies reported no serious side effects. Some small trials saw better walking or sleep scores. Larger reviews did not find clear benefits for blood sugar, cholesterol or muscle. The honest summary: NMN does what it says on the chemistry, and the health payoff is still being studied.

What NMN and NAD+ are

Every cell runs on chemical reactions. Many of them need a helper molecule called NAD+. In animal studies, NAD+ runs lower with age.

The body builds NAD+ from smaller pieces. NMN is one of the last pieces before NAD+. That is why scientists began giving NMN to people: if you supply the piece, does NAD+ go up?

What human trials found

These are randomized, placebo-controlled trials. That means some people got NMN, some got a look-alike pill, and nobody knew which.

Study Who and how long Daily NMN What it found What it did not find
Yi 2023 80 healthy middle-aged adults, 60 days 300, 600 or 900 mg Blood NAD rose in every NMN group. Walking distance improved more than placebo. No change in insulin resistance.
Igarashi 2022 Healthy older men, 6 or 12 weeks 250 mg Blood NAD+ rose. Well tolerated. Walking and grip changes were too weak to count. No change in body composition.
Yoshino 2021 Women after menopause with prediabetes, 10 weeks See paper Muscle insulin sensitivity rose. Only one specific group was tested.
Morifuji 2024 60 older adults, 12 weeks 250 mg Blood NAD+ rose. Faster 4-meter walk and better sleep scores than placebo. The main test (stepping) showed no difference.
Kim 2022 108 older adults, 12 weeks 250 mg Afternoon intake had the largest effect on a chair-stand test and drowsiness. Placebo groups improved on some measures too.
Liao 2021 48 recreational runners, 6 weeks 300, 600 or 1,200 mg Some oxygen-use measures improved at the two higher doses. No change in VO2max or peak power.
Katayoshi 2023 36 healthy middle-aged adults, 12 weeks 250 mg NAD+ markers rose. Well tolerated. Artery stiffness did not differ from placebo.
Akasaka 2023 14 older men with diabetes, 24 weeks 250 mg Safe over 24 weeks. No better grip or walking speed than placebo.

What we do not know yet

When researchers pool many trials, the picture gets more careful.

  • Blood sugar and cholesterol: a 2025 review of 12 trials (513 people) found NMN reliably raised NAD, but most health measures did not differ from control. Every trial in it had some risk of bias.
  • Muscle in adults over 60: a 2025 review found no clear effect on muscle mass, grip strength or walking speed.
  • Safety over years: a 2026 review of 15 trials found no rise in side effects or liver enzymes. But the longest trial ran 24 weeks.
  • Living longer: no human study has tested that. The lifespan results are from animals.

How to choose an NMN

NMN is a newer ingredient, and labels can be confusing. Four checks:

  1. Know the source. Look for a named, traceable NMN ingredient, not just "NMN powder."
  2. Read the NMN line, not the total. Some labels add NMN and other ingredients into one big number. Find the milligrams of NMN itself. The trials above used 250 to 1,200 mg a day.
  3. Skip hidden blends. A "proprietary blend" hides how much of each ingredient you get. Every ingredient should have its own amount.
  4. Ask about outside testing. A lab that is not the seller can check that the label matches what is in the capsule.

Questions readers ask

Is NMN safe?
Short trials in healthy adults reported no serious side effects. One study gave 1,250 mg a day for 4 weeks with no severe side effects, and another gave up to 900 mg a day for 60 days. Long-term studies do not exist yet.
How much NMN did the studies use?
Between 250 and 1,250 mg a day. Many trials used 250 mg.
Does NMN slow aging?
No human study has shown that. Human trials measured NAD+ levels, walking, sleep scores and blood tests over weeks, not lifespan.
Should I take it in the morning or afternoon?
One study found larger effects with afternoon intake, but it was a single trial. There is no settled answer.
Should I ask my doctor first?
Yes, especially if you take prescription medicine or have a health condition.
